In einem sich in einem Reagenzglas befindlichem Thioglykolat-Agar wird eine MO "eingestochen". Entlang des Einstichkanals bildet sich eine O2 Gradient. Aufgrund dem des Gradienten entsprechendem Wachstum kann auf die Sauerstofftolleranz des MO's geschlossen werden. So wachsen z.B. aerobe MO's oben und Anarobier unten.
